The plant 2S albumins exhibit a spectrum of biotechnologically exploitable functions. Among them, pumpkin 2S albumin has been shown to possess RNase and cell-free translational inhibitory activities. The present study investigated the anticancer, DNase and antifungal activities of pumpkin 2S albumin. The protein exhibited a strong anticancer activity toward breast cancer (MCF-7), ovarian teratocarcinoma (PA-1), prostate cancer (PC-3 and DU-145) and hepatocellular carcinoma (HepG2) cell lines. Acridine orange staining and DNA fragmentation studies indicated that cytotoxic effect of pumpkin 2S albumin is mediated through induction of apoptosis. Pumpkin 2S albumin showed DNase activity against both supercoiled and linear DNA and exerted antifungal activity against Fusarium oxysporum. Secondary structure analysis by CD showed that protein is highly stable up to 90 °C and retains its alpha helical structure. These results demonstrated that pumpkin 2S albumin is a multifunctional protein with host of potential biotechnology applications. 相似文献

A series of 3,6-disubstituted imidazo[1,2-b]pyridazine derivatives have been synthesized and characterized with spectroscopic analyses. The antifungal activities of these compounds against nine phytopathogenic fungi were evaluated by the mycelium growth rate method. The in vitro antifungal bioassays indicated that most of compounds displayed excellent and broad-spectrum antifungal activities. Especially, compounds 4a, 4c, 4d, 4l and 4r exhibited 1.9–25.5 fold more potent than the commercially available fungicide hymexazol against Corn Curvalaria Leaf Spot (CL), Alternaria alternate (AA), Pyricularia oryzae (PO) and Alternaria brassicae (AB) strains. Two new isoflavonoid glucosides, Achyranthosides A and B, were isolated from the roots of Achyranthes bidentata. Their structures were established through extensive analyses using 1H, 13C NMR, HSQC, HMBC, NOESY, and HREISMS spectroscopic data. The structures of the new compounds are characterized by the methoxyl groups substituted on the specific positions of the phenyl rings. The two compounds were evaluated for their anti-inflammatory activities on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW264.7 murine macrophage cells. A series of linear hydrophilic cationic polymers with different charge density and molecular weights were synthesized by one-step polymerization process. The effect of the hydrophobicity and molecular weights on the antifungal activity against Rhizoctonia solani (R. solani) and Fusarium oxysporum f. sp. cubense race 4 (Foc4) was assessed. The biotoxicity of the cationic polymers were evaluated based on their median lethal concentration (LC50) for zebrafish and silkworm and median lethal dose (LD50) for Kunming mice. The results indicated that the balance between antifungal activity and biotoxicity could be well tuned by controlling the hydrophobic-hydrophilic balance. The minimum inhibitory concentration (MIC) of PEPB10 and PEPB25 against R. solani were 160 μg/mL and 80 μg/mL, respectively. And the LD50 for Kunming mice of PEPB10 and PEPB25 were more than 5000 mg/kg, which mean that PEPB10 and PEPB25 with high hydrophilicity show low toxicity and better selectivity for R. solani. The cationic polymers can kill the R. solani by damaging their membranes and exchanging the Ca2+ or/and Mg2+ cations of their membranes or cell wall. These results help to understand the antifungal mechanism of low-toxic polymeric quaternary ammonium salts and highlight their potential application as highly selective fungicidal agents for controlling plant diseases. 相似文献

The carboxanilide systemic fungicide 2-iodobenzanilide (2-IB) after 2 h pretreatment at 0.25 m M inhibited K+ and SO4 2- uptake by excised corn roots ( Zea mays L., cv. Dekalb 342) up to ca 70 and 40%, respectively. Proton extrusion from corn roots was also reduced by ca 50% after 1 h contact, and the microsomal K+ -stimulated ATPase activity from corn roots and pea stems ( Pisum sativum L., cv. Alaska) inhibited by 50 and 72%, respectively. In contrast, the Mg2+ -ATPase activities of microsomes and mitochondria at pH 6.0 and 8.7, respectively, were unaffected. After 2 h of preincubation with 0.25 m M 2-IB, O2 consumption by corn roots and pea stems was inhibited by 12 and 18%, respectively. ATP content of corn roots was not altered by 2-IB treatment. Therefore, energy availability "in vivo" was unaffected and the primary effect on corn roots is suggested to be at the plasmalemma ATPase which forms the proton gradient.
With isolated pea stem mitochondria, 0.25 m M 2-IB inhibited O2 consumption by ca 60% when NADH or malate plus pyruvate were added as substrates; when succinate was used O2 consumption was unaffected. The mode of action on isolated mitochondria was different from that shown for carboxin and also formerly attributed to the whole class of carboxanilide fungicides. 相似文献

Lipopeptides are microbial surface active compounds produced by a wide variety of bacteria, fungi and yeast. They are characterized by highly structural diversity and have the ability to decrease the surface and interfacial tension at the surface and interface, respectively. Surfactin, iturin and fengycin of Bacillus subtilis are among the most studied lipopeptides. This review will present the main factors encountering lipopeptides production along with the techniques developed for their extraction and purification. The activities of superoxide dismutase and guaiacol-dependent peroxidase were studied in the ontogenesis of recessive homozygous
mutants ofArabidopsis thaliana Heynh.le-2 andnfz24, which are characterized by two- to threefold increases in tolerance to the herbicide norflurazone. The mutantsle-2 andnfz24 differed from the initial race Dijon in some phenotypic features, duration of ontogenetic stages, and dynamics of the superoxide
dismutase and peroxidase activities in ontogenesis. A single treatment of plants with norflurazone induced an accelerated
increase in the level of both enzymes in the mutants as compared to the wild type plants. Under the conditions of multiple
treatment with norflurazone, the mutantsle-2 andnfz24 displayed a higher tolerance to the bleaching effect of the herbicide and were characterized by a higher level of superoxide
dismutase. The data obtained suggest that the superoxide dismutase and peroxidase activities are controlled by both ontogenetic

The Caatinga is a semi‐arid domain, characterized by reduced humidity and high rates of anthropogenic impact. In addition to the low availability of water, carnivorous mammals are still exposed to a number of threats related to landscape modifications. We used data from camera traps and occupancy models to investigate the habitat use by carnivores in an area of Caatinga in northeastern Brazil. We found a negative correlation between the distance from wind farms and the occupancy probability of the jaguar, and a positive correlation with the occupancy probability of the jaguarundi. Puma and jaguarundi occupied primarily sites near watercourses, whereas the occupancy of the crab‐eating fox was correlated positively with the presence of poachers. The ocelot was detected more frequently at sites distant from human settlements, whereas the jaguar was detected more often in areas far from wind farms. We found a negative correlation between the distance of water and the detection of the ocelot. The detection of the crab‐eating fox was influenced positively by the detection of cattle. In addition to the negative influence of some anthropic activities, our results indicate that water is a very important resource for species, and the few permanent sources of this resource available in the area must be preserved. The replication of our research in other systems, worldwide, that are experiencing similar pressures, should permit a systematic evaluation of the management and conservation strategies needed to rebuild or maintain populations, restore ecosystems, and support conservation policies in human‐altered landscapes. A series of 3-amino- and polyaminosterol analogues of squalamine and trodusquemine were synthesized and evaluated for their in vitro antimicrobial properties against human pathogens. The activity was highly dependent on the structure of the different compounds involved and the best results were obtained with aminosterol derivatives 4b, 4e, 8b, 8e and 8n exhibiting minimum inhibitory concentrations (MICs) against yeasts, Gram positive and Gram negative bacteria at average concentrations of 3.12–12.5 μM. 相似文献
